AUSTIN, Texas – March 23, 2016 – Altogen Labs today highlighted the research applications of its in-house validated BT474 xenograft model, an established platform for HER2-positive breast cancer programs and for the study of acquired resistance to targeted therapy.
Published work using the BT474 model includes a 2015 OncoImmunology study by Zhang and colleagues investigating acquired resistance to trastuzumab and the development of novel HER2 antibodies. That work indicated that blocking the HER2 protein could trigger calreticulin exposure and subsequent tumor-specific T cell immunity, reducing tumor recurrence. Earlier work by van Slooten and colleagues in 1995 established BT474 as a model for hormone-dependent breast cancer and immune therapy, characterized by estrogen receptor positivity, c-ErbB-2 (HER2) overexpression, and an in vivo phenotype of poorly differentiated adenocarcinoma with high proliferative and turnover rate.
Resistance to targeted therapy is now the central problem in HER2-positive disease. Sponsors developing next-generation antibodies, antibody-drug conjugates, and combination regimens need a model with documented behavior in the resistance setting – not simply a HER2-positive line.
