Altogen Labs Bioluminescent Imaging Enables Real-Time Tracking of Orthotopic and Metastatic Tumor Growth

AUSTIN, Texas – August 1, 2023 – Altogen Labs today highlighted its in vivo bioluminescent imaging capability, which allows sponsors to track internal orthotopic and metastatic tumor growth in real time rather than relying on end-of-study tumor weight.

In a published Altogen Labs case study, a luciferase-expressing U87-Luc glioma cell line was used to monitor tumor progression in vivo. Bioluminescence was measured in a Night Owl imaging system (Berthold Technologies) ten minutes after intraperitoneal injection of luciferin, with luciferase expression quantified as emitted photons, giving both a visual image and a numerical measure of orthotopic tumor burden.

This matters for a structural reason. Subcutaneous flank tumors can be measured with calipers three times a week. Orthotopic and metastatic tumors cannot be palpated at all — and without imaging, a sponsor learns the outcome of the study only at necropsy, with no growth curve, no early efficacy signal, and no ability to adjust. Bioluminescent tracking converts a single endpoint into a longitudinal dataset.

Altogen Labs generates luciferase- and GFP-tagged cell line clones in-house, maintaining them under antibiotic selection so that only cells carrying the integrated vector survive, and validates the resulting model before deploying it in sponsor studies. Imaging integrates with biodistribution work, allowing delivery and efficacy to be assessed in the same animals.
