Liposome Encapsulation of Small Interfering RNA (siRNA) Molecules
Liposomes are composite structures made of phospholipids that are artificially made to encapsulate various types of nucleic acids within the aqueous core of the liposome. Due to the hydrophobic nature of the liposome outer membrane, it can fuse with other lipid bilayers, such as the cell membrane, and deliver its content via cellular fusion.
Lipofection is the process in which liposomes are used for transfection of DNA into a host cell. Liposomes are used for the highly efficient delivery of cargo molecules (DNA, RNA, and protein) into cells both in vitro and in vivo. Liposomes containing charged molecules and drugs can effectively deliver otherwise impermeable compounds within the host cell. This technology is known to be very useful for intracellular delivery of drugs and associated research and development.
Altogen Labs offers encapsulation of negatively charged molecules (e.g., siRNAs, shRNAs, microRNAs, mRNAs, plasmid DNAs, and proteins) into either one of several standard liposome formulations (PC : Cholesterol and DSPC : Cationic Lipid : PEGylated Lipoid : Cholesterol) or a custom liposome formulation. Custom experimental design of liposome is available to provide in vivo siRNA protection (in the liposomal core), PEG-modifications to reduce the innate immune response, and low cytotoxicity using biodegradable lipids and polymers.
Sonication, liposome extrusion, and microfluidization (microfluidics) methods allow the generation of liposomes of a defined size (from 50 nm to 400 nm). The customer should provide at least 0.25 mg of siRNA for liposome encapsulation. Liposome-encapsulated siRNAs are delivered to the customer in sterile vial to ensure consistency and eliminate potential contamination.
