Altogen Labs Study Demonstrates Successful Delivery of Nucleic Acids Across the Blood-Brain Barrier in Glioblastoma Models

AUSTIN, Texas – August 10, 2021 – A preclinical study performed by Altogen Labs has demonstrated functional delivery of small RNA and plasmid DNA across the blood-brain barrier into glioblastoma tissue, a result announced publicly in July 2021 and one of the more difficult delivery problems in neuro-oncology drug development.

The blood-brain barrier remains the principal obstacle to treating primary brain tumors. Compounds that perform well in vitro frequently fail in vivo simply because they never reach the tumor. Altogen Labs scientists designed and executed the in vivo work behind this announcement, developing and modifying the transfection protocol required to achieve efficient delivery into glioblastoma tissue in both spontaneous and chemically induced murine glioma syngeneic models.

The study demonstrated functional cargo delivery of negatively charged biomolecules — chemically modified siRNA and a 3.7 kb plasmid DNA construct — through the blood-brain barrier, together with high reproducibility across animals. Reproducibility is the metric that distinguishes a usable preclinical delivery platform from an isolated observation, and it is the standard Altogen Labs applies to every validated model in its portfolio.
