Altogen Labs MOLM-13 Leukemia Model Supports FLT3 Inhibitor Development in AML Research

AUSTIN, Texas – March 24, 2018 – Altogen Labs today detailed the research applications of its validated MOLM-13 xenograft model, a robust platform for evaluating FLT3 and HDAC inhibitors in acute myeloid leukemia.

The published record demonstrates the model’s utility in the resistance setting. A 2017 study used MOLM-13 to evaluate FF-10101, an irreversible FLT3 inhibitor developed specifically to overcome the mutation-based resistance observed with quizartinib, and reported potent, selective FLT3 inhibition even in the presence of resistance mutations. Work by Keegan and colleagues in 2014 evaluated the FLT3/CDK4 dual kinase inhibitor AMG 925 in the same model, finding STAT5/Rb-mediated reduction in xenograft tumor growth and the ability to overcome FLT3 mutation-mediated resistance.

FLT3-mutated AML remains one of the more difficult targets in hematologic oncology precisely because resistance emerges so reliably. Programs designed to address that resistance need a model where resistance biology has been characterized and where comparator data exists.

MOLM-13 has also been applied in the evaluation of HDAC inhibitors including pracinostat, vorinostat, and BPR1J-340. The model sits within an Altogen Labs leukemia and lymphoma collection that includes HL-60, K562, MOLT4, Daudi, DOHH2, Granta519, H929, KARPAS 299, OPM2, Raji, Ramos, RL NHL, and the A20 and EL4 syngeneic murine models.
