AUSTIN, Texas – December 14, 2021 – Altogen Labs today detailed its tetracycline-inducible RNAi knockdown cell line development service, giving researchers temporal control over gene silencing in target validation studies.
Constitutive knockdown has a well-known limitation. If the target gene is essential, cells carrying a stable knockdown construct either die or adapt, and the surviving population no longer represents the biology under investigation. Inducible systems solve this: the line is established and expanded with the target intact, then silencing is triggered on demand, allowing acute loss-of-function phenotypes to be observed cleanly.
Altogen Labs develops tetracycline-inducible knockdown lines alongside constitutive stable RNAi lines and stable overexpression lines, all delivered within the company’s 28-day stable cell line development timeline. Supporting capability includes PCR cloning and DNA vector construction for shRNA, vector design and construction services, and full validation of knockdown by qRT-PCR and Western blot.
Established lines can be banked under GLP-compliant conditions as master and working cell banks, preserving them for the duration of a program, and can be advanced into in vivo studies using the company’s validated xenograft models — allowing a target to be interrogated in cell culture and then in a living tumor with the same engineered material. CRISPR/Cas9 knockout and knock-in model development is available where permanent genomic modification is preferred to RNAi-mediated knockdown.
