AUSTIN, Texas – October 18, 2022 – An orthotopic glioblastoma xenograft model developed by Altogen Labs has been adopted as the reference in vivo platform for validating tissue-targeted nucleic acid delivery, as documented in published technical materials for nanoparticle-based in vivo transfection reagents.
In the referenced validation work, nanoparticle-conjugated RNA and DNA complexes were administered at constant pressure by tail vein injection into NOD/SCID mice carrying the Altogen Labs orthotopic glioblastoma model. Seventy-two hours after the first injection, brain and brain tumor tissues were homogenized and analyzed for target knockdown and protein expression, confirming delivery to tumor tissue.
The selection of an Altogen Labs model as a validation standard reflects what sponsors have relied on for more than fifteen years: models that behave consistently. Each model in the Altogen Labs collection is established through repeated tumor growth curves reproduced across multiple independent studies before it is offered to clients. A model that cannot reproduce its own growth kinetics cannot support a dosing decision, a go/no-go decision, or a regulatory submission.
Orthotopic implantation places tumor cells in the anatomically correct tissue environment, producing a more faithful representation of tumor biology, vascularization, and drug exposure than subcutaneous implantation alone. Altogen Labs offers orthotopic and metastatic configurations across brain, lung, breast, pancreatic, colorectal, liver, prostate, and other tumor types, with bioluminescent imaging available for real-time monitoring of internal tumor burden that would otherwise be unmeasurable until necropsy.
Study designs can incorporate intravenous, intratumoral, intraperitoneal, oral gavage, subcutaneous, intramuscular, intranasal, intratracheal, topical, and continuous infusion dosing, using micro-injection techniques and pump-controlled delivery.
