]]> Altogen Labs, a preclinical CRO laboratory, has announced the validation of a comprehensive array of 10 lung cancer xenograft models. These models are essential for testing in vivo efficacy of novel therapeutics against lung cancer, elevating the prospects for significant advances in the diagnosis, understanding, and treatment of primary lung carcinoma. Less than 20% of new lung cancer drugs that are effective in vitro are also effective in vivo (Cancer Cell, 2017). An array of lung cancer xenograft models can significantly improve the chances of a new lung cancer therapy being successful in humans, fewer therapies will fail in clinical trials, and more effective and less toxic lung cancer drugs will be developed. Altogen Labs offers several PDX models and 10 in-house validated lung cancer xenograft models, including A549, H460, H226, NCI-H1975, DMS273, LL/2, Calu-3, Calu-6, H1155, and NCI-H522. These xenograft models are derived from human lung cancer cells and are implanted into immunocompromised SCID mice (subcutaneous or orthotopic). These models are used to predict which therapies are most likely to be effective and safe in humans. As a result, fewer patients are exposed to ineffective or harmful therapies.

Lung cancer is the third most diagnosed cancer and is the leading cause of cancer deaths worldwide. In the U.S., only 17.4% of those with lung cancer live beyond five years, a rate lower than many other cancers. In preclinical research, CDX and PDX xenograft models stand as the benchmark for testing the effectiveness and safety of new lung cancer treatments. These studies are fundamental for evaluating novel pharmaceutical compounds in vivo, allowing researchers to study tumor behavior, screen new cancer therapies, and examine metastasis. Altogen Labs is a GLP compliant preclinical research laboratory with extensive experience in creating CDX and PDX xenografts, syngeneic / allograft models, and metastatic xenograft models. Our services are not limited to the cell lines listed above – we are a biology CRO company and we can use any commercially available animal model with any tumorigenic cell line. Our animal facility was established in 2009 and offers a number of IACUC regulated and GLP compliant preclinical research services. Our main focus is immunocompromised xenograft animal models (mouse). Most work is performed on contract basis and 100% of IP belong to the client.

We have worked with a number of tumorigenic cell lines and had very good results in terms of both tumor take as well as functional tumor growth inhibition by experimental compounds. Note that immunocompromised animals and associated care are expensive, requires approved IACUC protocol, veterinary services, certified barrier animal facility, clean room, and associated QA/QC. We offer both preclinical efficacy (xenograft) and safety (Pharm/Tox) studies. Both GLP and non-GLP services are available, however please note that the GLP services are ~2.5-3 times more expensive and takes ~3-4 times longer to complete. Altogen Labs has been offering preclinical research services for cancer research since 2009. ]]> AUSTIN, Texas – July 28, 2021 – Altogen Labs offers over 100 validated models of cell line-derived xenografts (CDX) and patient-derived xenografts (PDX), including syngeneic xenograft models. Xenograft IND testing services are performed to measure the activity and efficacy of novel anti-cancer compounds based on the rate of engrafted tumor growth. The most recent additions of xenograft models in 2021 consist of T47D (model of breast cancer), SKNAS (neuroblastoma xenograft), HL60 (leukemia model), and H460 (lung cancer xenograft model). Xenografting, or transplanting cells from one species (most commonly human tumor cells) into other species (immunocompromised laboratory mice), is an important method used in preclinical studies to research novel cancer treatment efficacy and safety. Compared to in vitro studies, xenotransplantation allows for more precision and accuracy in tumor representation and anti-cancer drug efficacies. Altogen Labs is a GLP-compliant laboratory that has extensive experience in creating human tumor xenografts, as well as provides preclinical safety and efficacy xenograft studies to the biopharma industry and cancer research centers worldwide.

]]> Sodium dichloroacetate (DCA) has been investigated as a novel cancer therapy since 2007 when Bonnet et al published research study demonstrating the efficacy of DCA in treating human cancers by inhibition of mitochondrial pyruvate dehydrogenase kinase (PDK). Altogen Labs, an Austin-based biotechnology and preclinical research company, performed an independent study exploring potential novel and accessible cancer treatment options based on structurally modified DCA analogues. The results of the study identified two cationic dichloric compounds, referred to as DCAH and DCMAH, as potential candidates for further investigation. The study, recently published in Cancer Biology & Therapy, demonstrated in vivo efficacy of structurally modified analogs (DCAH and DCMAH) in U87 human glioblastoma xenograft model. The anti-tumor effect is comparable to both DCA and the well-known chemotherapy drug bevacizumab used for treatment of glioblastoma.

Dr. Dmitriy Ovcharenko, Altogen Labs corresponding author of the article, described that this work was performed in collaboration with National Cancer Institute’s Developmental Therapeutics Program (NIH). Of the three primary characteristics varied in our test compounds (charge/polarity, electron distribution, and spatial arrangement), reversing the charge compared with DCA produced the largest effect on tumor viability. While these dichloric compounds were initially investigated due to their similarity to dichloroacetate (DCA), immunoblotting results suggested a cellular pathway effect for DCAH and DCMAH independent from that of DCA. According to Altogen Labs’ senior scientist Dr. Catrina Chitjian, this work suggests the clinical potential for DCAH and DCMAH as minimally toxic anti-cancer drugs and demonstrates the importance of additional studies. Interestingly, DCAH and DCMAH are simple small molecules traditionally used as building blocks for organic chemistry synthesis, which is promising for potentially inexpensive mass production.
