Altogen Labs extensive biotechnology and pharmacology research services include the following:

The customer sends the cell line(s) or compound of interest, and company scientists perform an apoptosis cell-based assay (> 70 cancer cell lines are available).
The relative amount of caspase per cell is estimated using luminogenic caspase-3 and caspase-7 substrates in a buffer system optimized for caspase activity, luciferase activity, and cell lysis.
Cells in logarithmic growth are seeded in 96-well plates in 0.2 mL media. Cells are cultured with a single concentration of compound of interest.
Cells are harvested at 6, 12, 24, and 48 hr after addition of compound.
The caspase reagent is added to the cells in the well, resulting in cell lysis, followed by caspase cleavage of the substrate and generation of a “glow-type” luminescent signal (produced by the luciferase reaction).
A luminometer is used to measure luminescence, and the signal generated is proportional to the amount of caspase activity present.
